| Summary, etc. | Background : Datura metel L is a frequently encountered plant species in the Solanaceae family, renowned for its beneficial medicinal characteristics Using naturally occurring bioactive compounds has been crucial in progressing contemporary medical practices The plant roots excrete a large number of organic compounds into the soil The rhizosphere, a thin soil zone around plant roots, is a hotspot for microbial activity, making it a crucial component of the soil ecosystem Secondary metabolites produced by rhizosphere bacteria have sparked significant curiosity in investigating their possible biological and pharmacological impacts Methods : Serial dilutions (10X) were prepared to isolate a variety of bacteria from the rhizosphere of Datura metel L. Bacterial isolates obtained from the rhizosphere of Datura metel L. were preliminary screened for antimicrobial activity against human bacterial and fungal pathogens The most potent bacterial isolates were identified through 16S rRNA gene sequencing analysis PCR screening was used to detect genes related to the biosynthesis of bioactive metabolites, including PKS, NRPS, and lipopeptide genes To verify the presence of these genes, the amplified fragments underwent sequencing This study outlined the propagation of bacterial strains and subsequent working up to extract potent pure secondary metabolites Chemical profiling of total bacterial extract was performed by LC Q-TOF/MS analysis The ethyl acetate extracts of the propagated culture media were then obtained and subjected to fractionation and purification utilizing various chromatographic techniques The structure elucidation of the purified compounds was accomplished through the utilization of various spectroscopic techniques, such as UV, MS, NMR (ID and 2D) and Raman spectroscopy Antimicrobial activities of pure compounds were evaluated against various human and phytopathogenic microorganisms using the agar well diffusion method, while MIC was determined using turbidity method Cytotoxicity was assessed using the MTT test on human malignant skin cells (A-431) compared with normal melanocytes (HFB 4), two breast cancer cell |

| Summary, etc. | مقدمة عامة يعتبر نبات الداتورا ميثيل من الأنواع النباتية الشائعة في العائلة الباذنجانية، وهو مشهور بخصائصه الطبية المفيدة و قد كان استخدام المركبات النشطة بيولوجيا التي تتواجد بشكل طبيعي أمزا بالغ الأهمية في تطوير الممارسات الطبية المعاصرة تفرز جذور النبات عددا كبيرا من المركبات العضوية في التربة و تعتبر منطقة الجذور، و هي منطقة تربة رقيقة حول جذور النبات نقطة حيوية للنشاط الميكروبي ، مما يجعلها مكونا أساسيا للنظام الديني للتربة و قد أثارت الإفرازات الثانوية التي تنتجها بكتيريا منطقة الجذور فضولا كبيرًا للتحقق من تأثيراتها الديولوجية و الدوانية المحتملة و قد ركزت هذه الدراسة على النقاط الأتية : تحضير التخفيفات المتسلسلة (X10) لعزل مجموعة متنوعة من البكتيريا من منطقة جذور نبات الداتورا ميابل فحص العزلات البكتيرية التي تم الحصول عليها مبدنيا بحثا عن نشاط مضاد للميكروبات ضد الكائنات الممرضة البشرية سواء بكثيرية أو فطرية تعريف البكتيريا الفعالة بيولوجيا من خلال تحليل التسلسل الجيني 1RNA 165 استخدام تفاعل البوليميراز المسلسل PCR للكشف عن الجينات المرتبطة بالتخليق الحيوي للنواتج النشطة بيولوجياء بما في ذلك جينات PKS و NRPS و lipopeptide. وللتحقق من وجود هذه الجينات ، خضعت الأجزاء المتضاعفة التحليل التسلسل تنمية السلالات البكتيرية بكميات كبيرة في الأوساط الغذائية الخاصة بها والعمل عليها بعد ذلك لاستخراج نواتج ثانوية نقية فعالة إجراء التعريف الكيميائي للمستخلص البكتيري الكلي عن طريق تحليل LC Q-TOF/MS الحصول على مستخلصات الإيثيل أسمينات من السلالات البكتيرية وإخضاعها للتجزئة والتنقية باستخدام تقنيات كروماتوغرافية مختلفة توضيح التركيب الخاص بالمركبات المنقاة من خلال استخدام تقنيات طيفية مختلفة، مثل الأشعة فوق البنفسجية و مطيافية الكتلة الطرفية ، و الرنين المغناطيسي النووي (أحادي وثنائي الأبعاد) و طيف رامان تقييم الأنشطة المضادة للميكروبات المركبات النقية ضد العديد من الكائنات الحية الدقيقة المسببة للأمراض البشرية و التنباتية باستخدام طريقة انتشار بار الأجار، في حين تم تحديد MIC باستخدام طريقة العكارة تقديم السمية الخلوية باستخدام اختبار MIT على عدد من الخلايا البشرية مثل خلايا الجلد السرطانية (431-A) مقارنة بالخلايا الصبغية الطبيعية 4 (HFB) ، و اثنين من خلايا سرطان الذي (7-MCF و 231-MDA-MB) ، و خلايا الكبد السرطانية (2-Hep) ، و خلايا سرطان القولون و المستقيم (116-HCT) مقارنة بالخلايا الظاهرية الطبيعية (THLE2) |
